Tesamorelin

Tesamorelin is a synthetic 44-amino-acid analog of endogenous growth hormone-releasing hormone (GHRH), modified with a trans-3-hexenoic acid group at the N-terminus to enhance metabolic stability and receptor binding affinity. It is the only GHRH analog to have received regulatory approval in its pharmaceutical form (FDA, 2010).Laboratory research has investigated Tesamorelin's interaction with the GHRH receptor (GHRH-R) on anterior pituitary somatotrope cells, examining downstream cAMP/PKA signaling, GH gene transcription (GH1), and pulsatile GH secretion patterns. In-vitro binding studies demonstrate preserved full agonist activity at the GHRH-R with improved proteolytic resistance compared to native GHRH(1-44).
